    Edge distributions of Ceutorhynchus assimilis and its parasitoid Trichomalus perfectus in a crop of winter oilseed rape (Brassica napus)

    The edge distributions of Ceutorhynchusassimilis and its parasitoid Trichomalusperfectus within a crop of winter oilseed rape were investigated between May and July using yellow water-traps arranged on a grid system. A randomisation test, which compared the observed arrangement of trap counts with permuted arrangements, was used to quantify the observed tendency of C. assimilisto congregate at the edges as it migrated into the crop; this tendency declined during the season. Trichomalus perfectus were distributed at the edge of the crop only during the early phase of its immigration in May, which occurred later than immigration of its host. It is considered that this simple means of quantifying insect spatial distribution could prove a useful tool in precision targeting of insecticides

    Clay minerals in delta deposits and organic preservation potential on Mars

    Clay-rich sedimentary deposits are often sites of organic matter preservation and have therefore been sought in Mars exploration. However, regional deposits of hydrous minerals, including phyllosilicates and sulphates are not typically associated with valley networks and layered sediments that provide geomorphic evidence of surface water transport on early Mars. The Compact Reconnaissance Imaging Spectrometer for Mars (CRISM) has recently identified phyllosilicates within three lake basins with fans or deltas that indicate sustained sediment deposition: Eberswalde crater Holden crater and Jezero crater. Here we use high-resolution data from the Mars Reconnaissance Orbiter (MRO) to identify clay-rich fluvial–lacustrine sediments within Jezero crater, which has a diameter of 45 km. The crater is an open lake basin on Mars with sedimentary deposits of hydrous minerals sourced from a smectite-rich catchment in the Nili Fossae region. We find that the two deltas and the lowest observed stratigraphic layer within the crater host iron–magnesium smectite clay. Jezero crater holds sediments that record multiple episodes of aqueous activity on early Mars. We suggest that this depositional setting and the smectite mineralogy make these deltaic deposits well suited for the sequestration and preservation of organic material

    Enhanced thylakoid photoprotection can increase yield and canopy radiation use efficiency in rice

    High sunlight can raise plant growth rates but can potentially cause cellular damage. The likelihood of deleterious effects is lowered by a sophisticated set of photoprotective mechanisms, one of the most important being the controlled dissipation of energy from chlorophyll within photosystem II (PSII) measured as non-photochemical quenching (NPQ). Although ubiquitous, the role of NPQ in plant productivity remains uncertain because it momentarily reduces the quantum efficiency of photosynthesis. Here we used plants overexpressing the gene encoding a central regulator of NPQ, the protein PsbS, within a major crop species (rice) to assess the effect of photoprotection at the whole canopy scale. We accounted for canopy light interception, to our knowledge for the first time in this context. We show that in comparison to wild-type plants, psbS overexpressors increased canopy radiation use efficiency and grain yield in fluctuating light, demonstrating that photoprotective mechanisms should be altered to improve rice crop productivity

    Single-Molecule Three-Color FRET with Both Negligible Spectral Overlap and Long Observation Time

    Full understanding of complex biological interactions frequently requires multi-color detection capability in doing single-molecule fluorescence resonance energy transfer (FRET) experiments. Existing single-molecule three-color FRET techniques, however, suffer from severe photobleaching of Alexa 488, or its alternative dyes, and have been limitedly used for kinetics studies. In this work, we developed a single-molecule three-color FRET technique based on the Cy3-Cy5-Cy7 dye trio, thus providing enhanced observation time and improved data quality. Because the absorption spectra of three fluorophores are well separated, real-time monitoring of three FRET efficiencies was possible by incorporating the alternating laser excitation (ALEX) technique both in confocal microscopy and in total-internal-reflection fluorescence (TIRF) microscopy

    Photoacclimation strategies in northeastern Atlantic seagrasses: Integrating responses across plant organizational levels

    Seagrasses live in highly variable light environments and adjust to these variations by expressing acclimatory responses at different plant organizational levels (meadow, shoot, leaf and chloroplast level). Yet, comparative studies, to identify species' strategies, and integration of the relative importance of photoacclimatory adjustments at different levels are still missing. The variation in photoacclimatory responses at the chloroplast and leaf level were studied along individual leaves of Cymodocea nodosa, Zostera marina and Z. noltei, including measurements of variable chlorophyll fluorescence, photosynthesis, photoprotective capacities, non-photochemical quenching and D1-protein repair, and assessments of variation in leaf anatomy and chloroplast distribution. Our results show that the slower-growing C. nodosa expressed rather limited physiological and biochemical adjustments in response to light availability, while both species of faster-growing Zostera showed high variability along the leaves. In contrast, the inverse pattern was found for leaf anatomical adjustments in response to light availability, which were more pronounced in C. nodosa. This integrative plant organizational level approach shows that seagrasses differ in their photoacclimatory strategies and that these are linked to the species' life history strategies, information that will be critical for predicting the responses of seagrasses to disturbances and to accordingly develop adequate management strategies.Fundacao para a Ciencia e Tecnologia (FCT), Portugal [PTDC/MAR-EST/4257/2014

    Achieving Self-Directed Integrated Cancer Aftercare (ASICA) in melanoma: protocol for a randomised patient-focused pilot trial of delivering the ASICA intervention as a means to earlier detection of recurrent and second primary melanoma.

    BACKGROUND: Melanoma is common; 15,906 people in the UK were diagnosed with melanoma in 2015 and incidence has increased fivefold in 30 years. Melanoma affects old and young people, with poor prognosis once metastatic. UK guidelines recommend people treated for cutaneous melanoma receive extended outpatient, hospital follow up to detect recurrence or new primaries. Such follow up of the growing population of melanoma survivors is burdensome for both individuals and health services. Follow up is important since approximately 20% of patients with early-stage melanoma experience a recurrence and 4-8% develop a new primary; the risk of either is highest in the first 5 years. Achieving Self-directed Integrated Cancer Aftercare (ASICA) is a digital intervention to increase total-skin-self-examination (TSSE) by people treated for melanoma, with usual follow up. METHODS: We aim to recruit 240 adults with a previous first-stage 0-2C primary cutaneous melanoma, from secondary care in North-East Scotland and the East of England. Participants will be randomised to receive the ASICA intervention (a tablet-based digital intervention to prompt and support TSSE) or control group (treatment as usual). Patient-reported and clinical data will be collected at baseline, including the modified Melanoma Worry Scale (MWS), the Hospital Anxiety and Depression Scale (HADs), the EuroQoL 5-dimension 5-level questionnaire (EQ-5D-5 L), and questions about TSSE practice, intentions, self-efficacy and planning. Participants will be followed up by postal questionnaire at 3, 6 and 12 months following randomization, along with a 12-month review of clinical data. The primary timepoint for outcome analyses will be12 months after randomisation. DISCUSSION: If the ASICA intervention improves the practice of TSSE in those affected by melanoma, this may lead to improved psychological well-being and earlier detection of recurrent and new primary melanoma. This could impact both patients and National Health Service (NHS) resources. This study will determine if a full-scale randomised controlled trial can be undertaken in the UK NHS to provide the high-quality evidence needed to determine the effectiveness of the intervention. ASICA is a pilot study evaluating the effectiveness of the practice of digitally supported TSSE in those affected by melanoma. TRIAL REGISTRATION: Clinical Trials.gov, NCT03328247 .
